چکیده

the Octet platform provides analytical methods for measuring antibody and protein concentrations and binding affinities through various stages of drug discovery and development (table 1). Octet systems circumvent many limitations of ELISA and HPLc assays, enabling informed decisions to be made earlier in both drug and process development – particularly when chromatography and other critical separation steps are needed. Octet systems perform automated sample analysis in 96and 384-well microplate formats. the simple Dip and ReadTM approach enables streamlined workflows and rapid quantitation of 96 samples in as little as 20 minutes, or 384 samples in 70 minutes. In a typical quantitation assay, biosensors coated with capture molecules dip into samples and the concentration of the target Rashi Takkar, Applications Scientist, [email protected]; Sriram Kumaraswamy, Director, Marketing Field Applications, [email protected] 10

Production and Characterization of Murine Monoclonal Antibodies Recognizing Conformational and Linear Epitopes Localized on Human IgA2 Molecules

Background: There are two subclasses of human IgA (IgA1 and IgA2) that differ in antigenic properties and in chemical composition. The constant domains of α1 and α2 heavy chains have >95% sequence homology though major structural differences exist in the hinge region. Quantitation of IgA subclass levels depends on the availability of monoclonal antibodies (MAbs) specific for conserved conformat...

MONOCLONAL ANTIBODIES TO HUMAN ALPHA-FETOPROTEIN

Alpha-fetoprotein (AFP), a serum glycoprotein belonging to the onco-developmental proteins group, serves as a marker both in cancer research and in studies concerning fetal development and fetal pathophysiology. Monoclonal anti-AFP antibodies are essential reagents in developing appropriate techniques for measurement of this protein. In this study, in order to produce anti-AFP monoclonal a...

Production and Characterization of Monoclonal Antibodies against Brucella abortus S (99) Surface Antigens

By immunizing mice with killed whole bacterial cells of Brucella abortus S (99), a panel of six hybridomas producing monoclonal antibodies (mAb) specific for the surface antigens of this bacterium were produced. ELISA was used to screen the hybridoma supernatants. Immunoblots of the cell extract indicated that three mAb were specific for S-LPS (Ba-1, Ba-2, Ba-3) and three others were reactive w...

Monoclonal Antibodies as Therapeutic Agents: Advances and Challenges

Despite the major advances in conventional forms of treatment (i.e. surgical techniques, radiotherapy and chemotherapy) and improved survival rates, cancer is still the second leading cause of death in developing countries. One major limitation of cytotoxic drugs and radiation in the treatment of cancer patients is their inability to discriminate between malignant and normal tissues. This in tu...

Antibody Response to Human Extracellular HER2 Subdomain Proteins in Mice

Background: In addition to passive immunotherapy using anti-HER2 monoclonal antibodies, active immunotherapy via HER2 targeting is an interesting approach to inducing specific anti-tumor immune responses. We have recently reported the immunogenicity of HER2 subdomains following DNA immunization and HER2 protein boosting. In the present study, we evaluated the immunogenicity of different HER2 ex...
